Technical Specifications
BD FACSCalibur – FL1 to FL4 Channel Dye Compatibility Reference
This reference table lists the four fluorescence detectors (FL1–FL4) on the BD FACSCalibur and commonly compatible fluorochromes from BD and other major vendors. Always verify filter configuration and compensation before finalizing panel design.
| Channel | Excitation Laser | Typical Emission Filter | Emission Range (Approx.) | Compatible Fluorochromes (BD + Other Vendors) |
|---|---|---|---|---|
| FL1 | 488 nm (Blue) | 530/30 | 515–545 nm | FITC, Alexa Fluor 488, BB515, GFP, CFSE, VioBright FITC, Zombie Green, Live/Dead Green, Calcein Green |
| FL2 | 488 nm (Blue) | 585/42 | 564–606 nm | PE, PE-Dazzle 594, PE-Texas Red (partial), PE-Alexa Fluor 610 (partial), Cy3, mCherry (partial) |
| FL3 | 488 nm (Blue) | 670 LP (Long Pass) | >650 nm | PerCP, PerCP-Cy5.5, PerCP-eFluor 710 (partial), PE-Cy5 (with caution), PE-Cy5.5 (partial) |
| FL4 | 635 nm (Red) | 661/16 | 653–669 nm | APC, Alexa Fluor 647, eFluor 660, Cy5, APC-H7 (partial spillover), Live/Dead Far Red |
Important notes:
- FL1–FL3 are excited by the 488 nm blue laser.
- FL4 is excited by the 635 nm red laser.
- PE-Cy5 may show significant spillover into FL4.
- The FACSCalibur is a 4-color analog instrument; careful compensation is critical.
- Always consult the Flow Core before purchasing tandem dyes.
