Sciex “QTRAP 6500+” mass spectrometer coupled to Shimadzu HPLC (Nexera X2 LC-30AD)

The Sciex “QTRAP 6500+” mass spectrometer, a hybrid triple quadrupole/ion trap with ESI source coupled to Shimadzu HPLC (Nexera X2 LC-30AD) is used for a relative and absolute quantification of metabolites. This instrument provides high sensitivity, a high linearity range (5-6 orders of magnitude), and excellent selectivity.

TripleTOF 6600 System

Sciex TripleTOF 6600 mass spectrometer coupled to Shimadzu HPLC (Nexera X2 LC-30AD)

The Sciex TripleTOF 6600, a high-resolution accurate mass spectrometer, offers high sensitivity, good dynamic range, robust quantitation, and fast scanning capability. The fast acquisition rate allows us to collect MS/MS data on every detectable analyte. This instrument is ideal for identification, quantitation, and discovery of metabolites in biological samples.

TOF-MS mode (Full scan) and Information Dependent Acquisition (IDA) mode (Product Ion scan) are utilized for collecting MS and MS/MS data respectively. The instrument is automatically calibrated using a calibration delivery system injected the APCI positive and negative calibration solution for every 5 samples. The system is controlled by Analyst TF 1.7.1 software (Sciex). Data are processed by Sciex OS software version 2.0.1, MarkerView, and XCMS software. The metabolites are identified with a high level of confidence using their retention time, exact mass and the match between sample MS/MS spectra (if detectable) with their standards in our "in house MS/MS library".
