"Regulating when and where a microtubule assembles: put a gamma-tubulin ring on it" Speaker: Tarun Kapoor, Ph.D. Professor at Rockefeller University Abstract: Microtubules, polymers of alpha/beta-tubulin, provide tracks for intracellular transport. These cytoskeletal filaments are required for cell division, regulation of cell polarity and morphogenesis. Microtubules are also targets for therapeutic agents used to treat diseases such as cancer. The formation of cellular microtubule networks is regulated by the γ-tubulin ring complex (γ-TuRC), a large (∼2.3 MD) multiprotein assembly. This complex not only regulates where and when a microtubule is formed, but may also control filament architecture, polarity and anchoring. I will discuss our cryo-EM reconstructions of the γ-TuRC and describe how it is assembled by >31 proteins, including γ-tubulin and an actin-containing "lumenal bridge". These structural models have guided the biochemical reconstitution of the γ-TuRC with 10 proteins and allowed st…
