Glycosylation of proteins and lipids is abundant, yet often overlooked. The Kohler research group is committed to developing and implementing new tools optimized for the study of glycosylated molecules. In particular, we developed photocrosslinking sugar analogs that can be metabolically incorporated into cellular glycoconjugates and used to covalently crosslink transient glycan-mediated interactions. We use these reagents to study the functions of two classes of glycoconjugates: (1) sialic acid-containing glycoconjugates (sialosides); and (2) O-GlcNAc-modified proteins. Using these unique reagents, we are tackling problems in fundamental biology, cancer metastasis, and infectious disease.
