Core Usage Request Form C: Cell Biology and Pathology

The objective of this core is to provide high quality histopathologic and scientific support for the research projects by localizing proteins and/or mRNAs in tissues and cultured cells and to offer pathology support. The technical expertise and equipment available in these laboratories are not available commercially and cannot be reproduced using facilities in individual laboratories of the investigators or existing campus cores at comparable costs. Drs. Moe and Richardson have an established history of expertise and competence in:

  1. Processing of tissues for routine paraffin embedding.
  2. Harvesting and flash-freezing of tissue for cryotomy.
  3. Performing detailed paraffin and cryo-microtomy.
  4. Performing standard histologic stains and special stains when required (Masson’s Trichrome for collagen, PAS for glycogen, Oil Red-0 for lipid and Von Kossa for calcium).
  5. Performing immunohistochemstry, lectin and enzyme histochemistry and in situ hybridization
  6. Maintenance of a library of wild type murine embryos and adult tissues embedded in paraffin for analysis of developmental gene expression by in situ hybridization.
  7. Bright-field, dark-field, phase-contrast, and epifluorescence microscopy and image analysis.
  8. Isolating tissue with laser microdissection.
  9. Confocal immunocytochemistry and single tubule immunohistochemistry

Services:

  • Light and Confocal Microscopy and Image Analysis

  • Training in the above procedures

Mode of operation:

  1. Investigators and their key personnel meet with Core Directors and staff before a project is launched.
  2. The Core Directors are actively involved in the experimental design of the various protocols.
  3. The Investigators determine the level of involvement from their laboratory. The Core can provide all the service or the investigator can work collaboratively with the Core.
  4. A mutually agreed plan will be formulated, and the estimated cost recovery and time line will be presented to the investigator.
  5. Plan further experiments.
  6. A feedback and quality control system will be employed for the users and the Core Directors to evaluate the success. This evaluation will in turn be monitored by the Executive Committee.
