Lipid Modification

Although it is still debating where the initial autophagosome membrane comes from, it is a consensus that PI3P generation is the first noticeable and essential change on the initiating membrane structures. The class III phosphatidylinositol 3-kinase (PI3KC3) has an evolutionarily conserved role in producing PI3P on autophagosome membrane. We are the first group to purify human PI3KC3 holocomplex, which contains seven stoichiometric subunits. Among these subunits, we identified Barkor (Beclin 1 interacting Autophagy Related Key regulator, also known as Atg14 or Atg14L (Like)) as the autophagy specific targeting and regulatory factor of PI3KC3.
